目的:探讨信号转导子和转录激活子1(STAT1)在大肠癌中的表达及其意义。方法:采用SYBRGreenⅠ荧光定量RT.PCR技术对50例大肠癌组织和相应正常大肠黏膜组织中STAT1mRNA的表达进行检测,并分析其表达情况与临床病理资料的相关性。结果:癌组织中STAT1mRNA的表达水平明显低于正常组织(P〈0.05);在合并淋巴结转移的癌组织中的表达水平亦低于无淋巴结转移的癌组织(P〈0.05);肿瘤TNM分期中,T3/T4期癌组织STAT1mRNA表达水平明显低于T1/T2期俨〈0.05)。结论:STAT1基因在大肠癌组织中低表达,可能与大肠癌的恶性程度相关.可作为大肠癌恶性潜能的检测指标之一。
Objective To investigate the expression and significance of signal transducer and transcription activator 1 (STAT1) in colorectal cancer. Methods The expression of STAT1 mRNA in 50 colorectal cancer tissues and 50 normal eolorectal tissues were determined by SYBR Green I quantitative real-time PCR, and its relation with clinicopathological parameters was analyzed. Results The expression of STAT1 mRNA was significantly lower in cancerous tissues than that in normal tissues (P 〈 0,05), was significantly lower in lymph node invasive group than that in non-lymph node invasive group (P 〈 0.05), and was significantly lower in stage T3/T4 cancer group than that in stage T1/T2 group (P 〈 0.05). Conclusion STAT1 is poorly expressed in coloreetal cancer tissues, and is highly correlated with invasion and metastasis, which may serve as a biomarker for invasiveness and metastasis of colorectal cancer.
