骨组织的结构是组织学教学的重要环节,骨组织切片是组织学实验和显微技术实验教学中不可缺少的教学内容。但是骨组织切片难度大、成本高,一直以来难以广泛应用于实验教学。在以往脱钙骨组织切片方法的基础上,应用冰冻切片方法,选择方便、易得的实验材料,使用合成胶水替代OCT(optimal cutting temperature compound)包埋剂,能够非常方便地控制骨组织材料的包埋位置和切片方向,采用滴染法染色,甘油封片后即可得到与传统脱钙骨组织切片方法效果相同的切片,从而使得骨组织切片方法成本降低,流程简化,极大降低了切片和染色的难度并减小所需时间,适合应用于教学实验。这实验设计对培养学生的学习兴趣,创新精神和动手能力发挥了重要的作用。
Learning the structure of bone tissue is important in histology, and bone tissue slides are significant in histology experiment and microscopical technique experiment. Since the bone tissue slicing procedure is costly and difficult to handle, it is not widely applied in experimental teaching. Based on the decalcified bone tissue section method, a new method is presented by improving the frozen section method. Instead of OCT( optimal cutting temperature compound) embedding medium glue is adopted. It can be very easy to control the embedding position of bone materials and the slicing direction. Then dropping is used to dye on slice. After sealing pieces by glycerin, slices which are as good as those get from traditional method can be obtained. This method could significantly cut the cost and simplify the process. This enables the process of slicing and staining to be finished fast, and is promising in experiment teaching. Altogether, the method plays an important role in improving the learning interest, innovation and practice ability of students.
