多房棘球绦虫重组BCG-Em14-3-3疫苗诱导BALB/c小鼠的保护力观察

目的探讨多房棘球绦虫(Em)重组BCG-Em14-3-3疫苗免疫后对受攻击感染BALB/c小鼠的囊重和抗体及其亚类的影响。方法将5×106克隆形成单位(CFU)和5×105CFU疫苗分别采用皮下注射和鼻腔内接种免疫BALB/c鼠1次,免疫后8周每鼠用50个Em原头节攻击感染,感染后18周剖杀小鼠,分离泡球蚴并称重,计算囊重抑制率;收集鼻腔内接种组鼠血清,测定IgG及其亚类和IgE水平。试验设有空载体、卡介苗(BCG)和磷酸盐缓冲液(PBS)对照组。结果疫苗皮下注射和鼻腔内接种组小鼠的囊重抑制率分别为82.35%和59.41%;疫苗鼻腔内接种组攻击后18周的鼠血清IgG、IgG2a、IgG2b和IgE水平均较接种前高(P〈0.05或P〈0.01),吸光度(A490)值分别为0.090±0.006、0.026±0.002、0.024±0.003和0.031±0.019,IgG1和IgG3水平低于接种前(P〈0.05或P〈0.01),A490值分别为0.042±0.011和0.156±0.004。结论多房棘球绦虫重组BCG-Em14-3-3疫苗可诱导小鼠产生较强的保护力,表现为血清IgG、IgG2a、IgG2b、IgE水平升高和棘球蚴生长受抑制。

英文摘要:

Objective To investigate the reduction of alveolar echinococcus weight and Ig level in mice immunized with recombinant BCG-Em14-3-3 vaccine of Echinococcus multilocularis(Em) and challenged by Em protoscoleces.Methods BALB/c mice were vaccinated by 5×106CFU subcutaneously and 5×105CFU intranasally respectively,with Blank vector,BCG and PBS served as controls.The mice were challenged with 50 Em protoscoleces in 8 weeks after vaccination and sacrificed in 18 weeks after infection.The weight of alveolar echinococcus was measured and reduction of the weight was obtained,sera in the intranasal group were gathered to examine the A490 value of IgE,IgG and its subclasses by ELISA.Results The alveolar echinococcus weight was reduced by 82.35% and 59.41% in the subcutaneous and intranasal immunization group,respectively.In comparison with before vaccination,the A490 value of IgG,IgG2a,IgG2b and IgE was 0.090±0.006,0.026±0.002,0.024±0.003 and 0.031±0.019 respectively in the 18 weeks after challenge of intranasal group with a signifcant increase,but that of IgG1 and IgG3 decreased,with a value of 0.042±0.011 and 0.156±0.004 respectively(P〈0.05 or P〈0.01).Conclusion rBCG-Em14-3-3 vaccine of Em may induce BALB/c mice to produce a strong protective immunity,in which IgG,IgG2a,IgG2b and IgE increased and the growth of alveolar echinococcus was inhibited.
