为了获得发酵生产柠檬酸中玉米粉原料预处理最佳的工艺条件,采用单因素试验和正交试验,系统分析了液化温度、液化pH、加酶量等因素对残渣淀粉含量、清液总糖含量、DE值、酶解效率等的影响。结果表明:随着液化温度的上升,DE值、残渣淀粉含量的变化呈现显著下降趋势,过滤速度呈现加快的趋势。95℃液化温度对改善DE值、过滤速度和残渣淀粉含量最有利,90℃时酶解效率最高。正交试验结果显示发酵产柠檬酸玉米原料预处理最佳条件为液化温度95℃、调浆pH 5.4、淀粉酶加量0.5 kg/t,该预处理条件能有效提高粮食利用率。
In order to obtain the optimal corn powder pretreatment conditions for citric acid fermentation,single factor test and orthogonal experiments were used to analyze the effects of liquefying temperature,liquefying pH and amylase addition on residual starch content,total sugar content,dextrose equivalent( DE) value and hydrolysis efficiency.Results indicated that liquefying temperature increment significantly decreased DE value,residual starch content and enhanced filtering rate.95 ℃ liquefying temperature was optimal for improving DE value,filtering rate and residual starch content.Hydrolysis efficiency was highest when liquefying temperature was 90 ℃. Orthogonal experiments results suggested that the optimized corn powder pretreatment conditions were 95 ℃ for liquefying,pH 5. 4 for pulp-mixing,and 0. 5 kg/t for amylase addition. The grain utilization rate could be improved significantly under the optimized pretreatment conditions.
