以灭活的嗜水气单胞菌为疫苗对亲鱼进行免疫后,分别检测了亲鱼自身及其胚胎中重要补体成分的含量以及替代途径补体活性。结果发现,母体免疫后亲鱼及其胚胎中补体成分C3和Bf的含量均显著升高,表明母体免疫能够促进母源性补体的垂直传递。补体活性的测定结果发现,亲鱼免疫后其自身和胚胎中的替代途径补体活性也相应升高,表明由亲鱼传递给子代的母源性补体具有功能活性,有助于提高子代的抗感染能力。此外,还发现随着亲鱼体内补体含量及活性的升高,其胚胎中相应成分的含量与活性也随之升高,而且免疫2周后亲鱼胚胎体内补体含量及其活性的升高最为显著,表明提高亲鱼体内补体成分的含量能够促进母源性补体的垂直传递。由此推测亲鱼免疫是提高子代免疫力的有效措施。
This study was conducted to immunize the maternal Danio rerio with the formalin-killed Aero- monas hydrophila ,and then determin the contents of complement component C3 and Bf and the alternative complement activities (ACHs0) in both the mother and the embryos from them. Results showed that ma- ternal immunization of female Danio rerio caused a significant increase in C3 and Bf contents in the mother and a corresponding rise in the offsprings, which showed that the immunization of the parent fish could stimulate the vertical transfer of maternal complement to offsprings. Moreover, maternal immunization also induced a remarkable increase in the ACHe0 in both the mother and offspring, suggesting that the immune activity of maternal complement could also been transferred, so as to protect the embryos against pathogen- ic attacks. It was also found that the complement content and activity increased in response to the increase of complement content and activity in maternal fishes,and the complement content and activity in mother and the offspring both peaked in 2 weeks after maternal immunization. Therefore,immunization of the ma- ternal fishes provide an effective way to improve the immunity of their offsprings.
