以杜仲嫩叶为外植体,诱导愈伤组织并进行增殖试验。在培养基中添加不同浓度的Cu2+研究Cu2+对杜仲愈伤组织诱导及增殖的影响。结果表明:诱导培养基中附加1.0mg/L的Cu2+时,有利于杜仲愈伤组织形成,诱导率最高。在增殖培养基中添加0.7mg/L的Cu2+时,可明显改善愈伤组织的生长状态,增加愈伤组织增殖生长量。杜仲愈伤组织诱导率及增殖生长量随着Cu2+浓度的升高而逐渐升高,但当Cu2+浓度超过1.0mg/L时,诱导率及生长量下降。
The leaves of E. ulmoides Olive were taken as explants and induced callus formation and proliferation. On the medium,the different concentrations of Cu2+ were supplemented into the media to study the effects on induction and proliferation of E. ulmoides Olive callus. The results showed that Cu2+ was advantageous to callus information , and the induction rates were the highest on the induction medium contained 1.0 mg/L Cu2+. Add 0.7 mg/L Cu2+ in value-added medium could proliferation growth quantity. The rates of induction and proliferation increased gradually along with the increasing of the concentrations of Cu2+. But the critical Cu2+ eontent was 1.0 mg/L.
