目的探讨摄入贵州某品牌38°食用白酒后,不同剂量和不同时长对血清睾酮水平及大鼠睾丸间质细胞内类固醇激素急性调节蛋白(StAR)、表皮生长因子(EGF)、嗜铬颗粒素A(CgA)表达的影响,为科学健康饮酒提供基础实验依据。方法健康成年雄性SD大鼠69只,随机分为正常对照组(n=15)和实验组(n=54)。实验组随机分为4周组、8周组、12周组,每组18只大鼠,每个时间点又分为3个剂量组,即低剂量组、中剂量组、高剂量组。实验组大鼠每天灌胃2次,正常对照组正常喂养。分别在第4、8、12周末,采血清,取睾丸组织。采用免疫组化SABC法、图像分析、蛋白免疫印迹、化学发光法进行研究。结果各实验组的血清睾酮水平与正常对照组比较未见明显异常,差异无统计学意义(P〉0.05)。与正常对照组相比,4周、8周、12周各实验组StAR的表达有所增强(P〈0.05);4周、8周、12周各实验组EGF、CgA的表达无明显差异(P〉0.05)。结论在本实验设定的剂量和时程内用该白酒灌胃后,血清睾酮水平并无明显变化,大鼠睾丸间质细胞内StAR表达水平有所增强,EGF、CgA的表达水平无明显变化。
Objective To investigate a brand of 38 - degrees of edible alcohol from Guizhou on rat Leydig cells in steroidogenic acute regulatory protein (STAR) , epidermal growth factor (EGF), chromogranin A (CgA) expression and serum testosterone. Methods Sixty normal adult male SD rats were randomly divided into normal control group (n = 15) and experimental groups ( n = 54 ). Experimental groups included low dose group, mediate dose group and high dose group. The rats in experimental groups were given with corresponding dose of liquor gavage twice a day. At the end of the 4th, 8th and 12th week, the serums were collected and testicular tissue was taken. Immunohistochemical SABC method, image analysis, Western - blotting, chemiluminescence method were applied. Results There was no significant difference in serum T among all 4 groups (P 〉 0. 05 ). Compared with the normal control group, the expression of StAR was significantly up - regulated in all 3 experimental groups (P 〈 0. 05 ). Conclusion No serum testosterone alteration is observed in experiment groups, nor is the expression of EGF or CgA. StAR expression is enhanced by liquor intake.
