OSAS大鼠模型的实验研究

目的通过咽腔多点注射透明质酸钠凝胶建立大鼠阻塞性睡眠呼吸暂停综合征(obstructive sleep apnea syndrome,OSAS)模型。方法在Wistar大鼠咽腔多点注射透明质酸钠凝胶建立OSAS模型,监测OSAS大鼠脑电、口鼻气流及动态血氧2 h,平均血氧饱和度、最低血氧饱和度及睡眠呼吸暂停指数造模前后统计有差异判定模型成功。HE染色法观察大鼠皮质及海马结构。结果①OSAS大鼠平均血氧饱和度(89.75±2.01)、最低血氧饱和度(72.77±5.19)降低,睡眠呼吸暂停指数(5.70±1.02)增高,与造模前比较有统计学差异(P〈0.01)。②OSAS组大鼠脑组织HE染色皮质及海马区有神经元缺失、排列紊乱及脑小血管增生。结论①采用大鼠咽腔多点注射透明质酸钠凝胶,可成功制作大鼠OSAS模型。②OSAS大鼠的血氧、呼吸暂停指数及皮质海马区神经元的形态皆发生变化。

英文摘要:

Objective To set up rat obstructive sleep apnea syndrome(OSAS) model using the pharynx multi-points injection of sodium hyaluronate gel.Methods The pharynx multi-points injection of sodium hyaluronate gel in Wistar rats was applied to establish OSAS model,The EEG,oronasal airflow,and dynamic oxygen for 2 hours in OSAS rats were measured.The differences in the mean oxygen saturation,lowest oxygen saturation and sleep apnea index were determined to evaluate the model success.HE staining was performed to observe rat cortex and hippocampus structures.Results ① The mean oxygen saturation(89.75 ± 2.01) and lowest oxygen saturation(72.77 ± 5.19) in OSAS rats were reduced,while sleep apnea index(5.70 ± 1.02) was increased.There were statistically significant differences compared with those before model establishment(P 0.01).② HE staining showed that the neuronal loss,disorganization and small vessel proliferation were discerned in OSAS rat cortex and hippocampus.Conclusion ① Rat OSAS model could be successfully established through the pharynx multi-points injection of sodium hyaluronate gel;② Blood oxygen,sleep apnea index and the neuronal morphology in the cortex and hippocampus were changed in OSAS rats.
