为了探究天麻蜜环菌粉正己烷提取物抗炎作用的物质基础及抗炎机制,本文首先对天麻蜜环菌粉正己 烷提取物进行薄层层析和中低压柱层析分离,获得了具有显著抗炎活性的组分5 ( Fr. 5 ),进一步通过硅胶柱分 离,得到主成分较为集中的组分5-l(Fr.5-l), 液质联用( L C - M S )及高效液相色谱分析推测F r . 5 - 1的主成分为大 豆黄素;其次,通过脂多糖( LPS)诱导小胶质细胞( B V 2 )炎症模型考察F r . 5 - 1不同剂量( 1 0、 3 0、100 p g / m L )对 炎症因子-氧化氮( NO)和肿瘤坏死因子( TNF-ct)表达的影响,发现中高剂量( 3 0、100 |xg / m L )的 F r . 5 - 1能显著 抑制 LPS诱导的BV2细胞炎症因子NO 及 T N F - c t的分泌.研究表明天麻蜜环菌粉提取物Fr. 5 - 1具有抗炎活 性,其机制可能是通过抑制炎症因子的表达和分泌.
In order to explore the material basis and anti-inflammatory mechanism of n-hexane extract of Armillaria-mellea firstly,we identified fraction Fr. 5 by TLC and column chromatography separation in low pressure. Then this frac-tion was chromatographed on a silica gel column to get Fr. 5-1. The main component of Fr. 5-1 may be compound Daid- zein by liquid chromatography-mass spectrometry( LC-MS) and HPLC analysis. Fr. 5-1 (30,100 jjig/mL) significantly inhibited LPS induced NO and TNF-a secretion in microgliacells ( BV2) . Therefore, the result demonstrated that Fr. 5-1 from A. mellea had anti-inflammation activity in LPS activated microglia through inhibition of inflammatory mediators.
