目的:观察艾炷灸对兔动脉粥样硬化(AS)斑块中过氧化酶体增殖物激活型受体γ(PPARγ)及基质金属蛋白酶-9mRNA(MMP-9mRNA)表达的影响,探讨艾炷灸对兔动脉粥样硬化的形成及斑块稳定性的影响。方法:通过高脂饲料喂养及免疫损伤法建立兔AS模型。将60只新西兰大耳白兔随机分为4组,每组15只,即空白组,模型组,艾炷灸组,西药组。治疗16周后检测兔粥样硬化斑块中PPARγ及MMP-9mRNA的表达。结果:与模型组比较,艾炷灸组和西药组的PPAR1平均灰度值显著降低(P〈0.05,P〈0.01);艾炷灸组和西药组的MMP-9mRNAIA值明显低于模型组(P〈0.05,P〈0.01);艾炷灸组与西药组比较,MMP-9mRNAIA值差异有统计学意义(P〈0.01)。结论:艾炷灸可以通过激活PPARγ的蛋白表达、抑制MMP-9mRNA的表达,对AS主动脉血管内皮细胞产生-定的保护作用,但对抑制MMP-9mRNA的表达作用,西药组疗效优于艾炷灸组。
Objective: To observe the effects of direct moxibustion on the expression of peroxisome proliferator-activated receptor γ(PPARγ) and matrix metallop-roteinases-9 mRNA (MMP-9mRNA)in atherosclerotic-plaques of atherosclerosis (AS) rabbits and to explore the influence of moxibustion on formation and stability of atherosclerotic-plaque of rabbit. Methods: AS model of rabbit was established by using high-fat diet combined with immunologic injury. 60 New Zealand white rabbits were divided into 4 groups: blank group (control group), model group (AS model group), direct moxibustion group (AS model plus direct moxibustion) and western medicine group (AS model plus atorvastatin), 15 rabbits in per group. The expressions of PPARγ and MMP-9 mRNA in atherosclerotic plaque of rabbits were measured after intervening for 16 weeks. Results: Compared with the model group, expressions of PPARy in western medicine group and direct moxibustion group were higher with significant difference (P〈0.05 or P〈0.01). Compared with the model group, expressions of MMP-9 mRNA in direct moxibustion group and western medicine group were lower with significant difference (P〈0.05 or P〈0.01). There were significant differences in the comparison of MMP-9 mRNA expression between the direct moxibustion group and western medicine group (P〈0.01). Conclusion: Direct moxibustion can activate the protein expression of PPART, and inhibit the expression of MMP-9 mRNA and have certain protective effects on aortic endothelial cells of AS rabbits. But for the effects of inhibiting the expression of MMP-9 mRNA, western medicine group is better than direct moxibustion group.
