目的:观察隔药饼灸对结肠5-HT及中枢c-fos表达的影响,探讨隔药饼灸对功能性胃肠病(FGIDs)肝郁脾虚证大鼠治疗作用的可能机制。方法:将48只SD大鼠按随机数字表法随机分为4组,即:空白组、模型组、隔药饼灸组和逍遥散组,每组12只。空白组正常饲养,不做任何处理;余3组均采用复合病因造模法(慢性束缚应激+过度疲劳+饮食失节+夹尾+摇晃)连续21d造模后,模型组行捆绑束缚30min,并灌服0.9%氯化钠溶液,灌胃容积均为1mL/100g体质量(下同);隔药饼灸组行隔药饼灸30min,并灌服0.9%氯化钠溶液;逍遥散组行捆绑束缚30min,灌服逍遥散,连续干预14d。实验结束后采用高效液相检测结肠5-HT表达,荧光定量PCR法检测海马、杏仁核c-fos表达。结果:与空白组相比,模型组大鼠结肠组织中的5-HT的表达明显增加(P〈0.01);与模型组相比,逍遥散组和隔药饼灸组结肠组织中5-HT表达下降(P〈0.01,P〈0.05);且隔药饼灸组优于逍遥散组(P〈0.05)。与空白组相比,模型组c-fos表达均增多(P〈0.05);与模型组相比,隔药饼灸组和逍遥散组c-fos表达减少(P〈0.05);隔药饼灸组优于逍遥散组(P〈0.05)。结论:隔药饼灸能下调结肠组织中5-HT表达来改善胃肠吸收和感觉异常,结肠5-HT及中枢海马、杏仁核c-fos异常表达提示FGIDs发病机制不仅与内分泌免疫系统方面有关,同时神经系统也参与该病发生,提示脑肠轴-中枢途径可能是隔药饼灸治疗FGIDs关键所在。
Objective: To observe the influence of cake-separated moxibustion on the expression of 5-HT and central c-los in rats with functional gastrointestinal disorders (FGIDs), and to investigate the possible mechanism and therapeutic effect of cake-separated moxibustion in rats with FGIDs of liver-depression and spleen-deficiency syndrome. Methods: 48 SD rats were divided into 4 groups according to the table of random number: a blank group (A), a model group (B), cake-separated moxibustion group (C), Xiaoyao Powder group (D), 12 in each group. Rats in group A were normal feeding without any treatment. The rest three groups were made with complex modeling methods for 21 days (chronic restraint stress + excessive fatigue + irregular food + tail clamp + shake). Group B was bounded for 30min and given normal saline by gavage, and the gavage volume was 1mL/100g (the same below). Group C was treated with cake-separated moxibustion for 30min, and given normal saline by gavage. Group D was bounded for 30min, and given Xiaoyao Powder by gavage for 14 days. After the experiment finished, the expression of 5-HT in colon tissues was detected by high performance liquid chromatography (HPLC), and the expression of c-fos in hippocampus and amygdala were detected by fluorescence quantitative PCR. Results: Compared with the blank group, the expression of 5-HT in colon tissues of rats in the model group was statistically significantly different (P〈0.01). Compared with the model group, the expression of 5-HT in colon tissue of rats decreased in Xiaoyao Powder group and cake-separated moxibustion group, and the differences were statistically significant (P〈0.05, P〈0.01). Compared with Xiaoyao Powder group, the difference in cake-separated moxibustion group was statistically significance (P〈0.05).
