为了研究羊口疮病毒(ORFv)新疆野毒株的遗传进化情况,参照GenBank公布的ORFvB2L和FII.基因序列,设计特异性引物,对CIRFvSHZ1新疆野毒株B2L和F1L基因进行PCR扩增,克隆、测序后进行遗传进化分析。结果表明:新疆野毒株SHZ1与GenBank公布的不同地域的17个流行株相比,B2L基因核苷酸同源性为97.28%~98.86%,推导氨基酸同源性为88.65%~90.50%;F1L基因核苷酸同源性为94.56%~97.07%。推导氨基酸同源性为93.82%~95.88%。基于B2L基因的遗传进化树分析发现,SHZ1株B2L基因与我国现用疫苗株B2L基因亲缘关系较近;而基于F1L基医的遗传进化树显示,SHZ1株与意大利分离株的亲缘关系最近。本研究提示0RFv SHZ1株可能是国外流行株与疫苗株重组后产生的一个变异株。
In order to investigate the genetic evolution situation of Orfv wild strain,specific primers were designed according to the nucleotide sequence published by GenBank and used to amplify Orfv B2L gene and F1L gene. Then the amplified B2L gene and F1L gene of Xinjiang wild strain SHZ1 were sequenced for the phylogenetic analysis. We compared the Xinjiang wild strain with other seventeen wild strains published by GenBank, and found that nucleotide identities of B21 genes was 97. 28%- 98.86% ;derived amino acid identities was 88.65%-90.50% ;nucleotide identities of F1L gene was 94.56%-97.07% ,derived a mino acid identities was 93.82%-95.88%. Based on the analysis of the B2L gene, genetic evolutionary tree showed that the SHZ1 strain was closer to the current domestic vaccine strains. However, the genetic evolutionary tree based on F1L gene showed that the SHZ1 strain is the closest to the isolate from Italy. It is inferred that Orfv wild strain SHZ1 is probably a variation strain which is occurred through the recombination between foreign prevalent strain and domestic vaccine strain.
