建立动物疫病的快速诊断,对于我国养殖业疫病的预防及控制、进出口检疫、动物食品安全卫生等方面具有重大意义。重组酶聚合酶扩增技术(RPA)是近几年出现的1种有望替代PCR的新的核酸扩增技术。该技术主要依赖于3种酶:能结合单链寡核苷酸引物的重组酶、单链DNA结合蛋白和DNA聚合酶。反应过程中不需要模板链的热变性,可以在恒定的低温条件下快速扩增DNA或者RNA。具有特异性强、灵敏度高、快速、操作简便、适用于现场快速诊断等特点,在动物疾病早期诊断、实地检测、进出口快速检疫等方面具有巨大的应用前景和市场。本文对RPA技术进行综述,以期为相关研究提供参考。
Establishing a rapid diagnostic method for animal disease will be benefit for the preven- tion and control of livestock disease, the quarantine of import and export, and the safety of animal foodstuff. Recombinase polymerase amplification (RPA) is a newly developed nucleic acid amplifi- cation technology that is higher efficient and more suitable in filed detection than polymerase chain reaction method (PCR). Three enzymes are needed in RPA technology,including recombinant cn zyme,single-stranded DNA binding protein and DNA polymerase. Unlike PCR, heat denaturation of the template strand is not needed in the RPA technology. Besides, RPA is proved to be high spe cific,sensitive,fast,and easy to be operated,therefore,it is more suitable for quick field diagnosis than other technologies in the early diagnosis of animal diseases, rapid quarantine of import and ex- port. Here we summarized the newly development and application of RPA technology to provide valuable reference for related research.
