目的:观察解毒活血法对肾间质纤维化大鼠TGF-β1的影响,探讨其抑制肾间质纤维化的作用机制。方法:SD大鼠随机分为假手术组,模型组,缬沙坦组,解毒活血低剂量组,解毒活血高剂量组,每组10只。除假手术组,其余各组大鼠结扎左侧输尿管复制肾间质纤维化模型。所有大鼠灌胃给药,缬沙坦组给予26 mg/kg;解毒活血低剂量组予解毒活血中药12g生药/kg;解毒活血高剂量组予24g生药/kg,假手术组和模型组予等容量生理盐水,共14天。左肾组织行HE、Masson染色,观察病理形态学改变,免疫组化检测肾组织中TGF-β1的蛋白表达。结果:模型组大鼠间质大量炎症细胞浸润,胶原成分明显增多,TGF-β1呈强阳性表达,各治疗组炎症细胞浸润及胶原沉积程度均比模型组显著减轻,TGF-β1表达明显减弱。结论:解毒活血方药可抑制肾组织中TGF-β1的表达,继而减少细胞外基质沉积,延缓肾间质纤维化进程。
Objective:To observe the effects of Jieduhuoxue therapy on the expression of TGF-β1 in UUO rats,and to investigate its mechanism in renal interstitial fibrosis.Methods:Fifty Sprague-Dawley rats were randomized equally into 5 groups:Control group,UUO group,Valsartan group,low-dose of Jieduhuoxue decoction group and high-dose of Jieduhuoxue decoction group.Except the Control group,rats in other four groups were made into renal interstitial fibrosis model by ligaturing the left ureter.All rats were treated with drugs by oral infusion once a day.Rats in Valsartan group,Jieduhuoxue decoction low-dose group and high-dose group were treated respectively with Valsartan 26mg/kg,Jieduhuoxue decoction 12g/kg and 24g/kg for fourteen days.The protein expressions of TGF-β1 in rats' kidney were detected by immunohistochemical.Results:Compared with control group,the inflammatory cells infiltration and collagen deposition of the model group in renal interstitial space increased significantly,the expression of TGF-β1 was enhanced.After treatment,the inflammatory cells infiltration and collagen deposition decreased,the high expression of TGF-β1 was down-regulated.Conclusion:Jieduhuoxue decoction has the effect of inhibiting the over expression of TGF-β1,reducing the deposition of extracelluar matrix,delaying the procedure of renal interstitial fibrosis.
