目的探讨COPD患者的病程与气道上皮细胞分泌PGE_2和TGF-β_1的改变情况。方法将本院呼吸科门诊及住院部61例COPD患者按COPD诊断标准分为稳定期组43例,急性发作期组18例,并取本院体检正常者20例为对照组。用纤维支气管镜刷取气道上皮细胞后进行体外分离和培养。48 h后用ELISA试剂盒测定其上清液中PGE_2和TGF-β_1分泌水平。结果三组中PGE_2(ng/L)在对照组为(102.05±12.2.3)、稳定期组为(142.17±18.05)、急性加重期组为(195.37±20.76)。TGF-β_1(ng/L)在上述三组中依次为(0.36±0.08)、(0.54±0.11)和(1.32±0.43),三组间均有显著性差异(P〈0.05)。结论 COPD病情的发作与气道上皮细胞分泌PGE_2和TGF-β_1加强之间有一定关联。
Objective To investigate the change of airway epithelial cells secreted PGE_2 and TGF-β_1in different stages of COPD patients. Methods Sixty-one of patients with COPD in our hospital were divided into stable group(43 cases) and acute exacerbation group(18 cases) according to the COPD diagnostic criteria. And 20 cases of healthy person was attributed to the control group. The airway epithelial cells were isolated and cultured in vitro with the brush of the fiber bronchoscopy. After 48 h, the levels of PGE_2 and TGF-β_1in the supernatant were determined by ELISA kit. Results Among the three groups, PGE_2is(102.05±12.23) ng/L in the control group,(142.17 ±18.05) ng/L in stable group, and(195.37±20.76) ng/L in acute exacerbation group, and TGF-β_1in the three groups are(0.36 ±0.08) ng/L,(0.54±0.11) ng/L,(1.32±0.43) ng/L respectively, which have significant difference(P〈0.05). Conclusion There is a certain correlation between the attack of COPD and the secretion of PGE_2 and TGF-β_1with airway epithelial cells.
