以厦门树白蚁(Glyptotennes Xiamenensis Li et Huang)为原料,经PBS缓冲液(pH6.7)抽提、硫酸铵分级分离获得纤维素酶粗提物(简称EGase),经DEAE-Sepharose Fast Flow阴离子交换柱层析、Shephadex G-100凝胶过滤柱层析等纯化,得到电泳单一纯的酶制剂,比活力为458.7U/mg.研究了金属离子对该酶活力的影响:结果表明,正-价金属离子Li^+、Na^+、K^+对酶活力没有影响,Ca^2+、Zn^2+和Mn^2+有激活作用,重金属离子Cu^2+、Pb^2+和Hg^2+有抑制作用,表现为不可逆抑制.
An endo-1,4-β-D-glucanase (EC#3.2.1.4)was purified from the termite Glyptotennes Xiamenensis Li et Huang by PBS buffer ( pH 6.7 ) extraction, ammonium sulfate precipitation, the ion-exchange chromatography on DEAE-Sepharose Fast Flow and the gel filtration through Sephadex G-100. The final preparation was homogeneous and electrophoretic pure with the specific activity of 458.7 U/mg. The effects of several metal ions on the enzyme activity were studied. Li^+, Na^+, K^+ had no effects on the enzyme activity. The enzyme activity was stimulated by Ca^2+ , Zn^2+ and Mn^2+ but inhibited by Cu^2+ , Pb^2+ , Hg^2+ following the irreversible mechanism.
