为了了解环境因子对玫烟色棒束孢分生孢子萌发的影响,采用凹玻片悬滴等方法研究了温度、湿度、光周期、初始pH值、紫外照射、高温胁迫及吐温-80浓度对玫烟色棒束孢IF-1106菌株孢子萌发的影响。结果表明:玫烟色棒束孢孢子萌发的最适温度为26℃左右,适温为23~32℃,玫烟色棒束孢分生孢子萌发对湿度的要求较高,低于85%分生孢子不萌发。黑暗促进孢子萌发,光照抑制孢子萌发,但光暗交替时,随着光照时间的延长,孢子萌发率升高。孢子萌发最适pH值为5~7,用0.05%~1%的吐温-80配制孢子悬浮液。经过48℃高温和紫外线照射处理后,玫烟色棒束孢孢子的萌发都受到很大程度的抑制。因此玫烟色棒束孢IF-1106菌株分生孢子萌发最适条件为温度26℃,相对湿度95%以上,初始pH值为6,全黑暗培养。
In order to understand the effects of environmental factors on conidia germination of Isaria fumosorosea1, concave glass hanging drop and other method were used to study the effects of temperature, humidity, photoperiod, initial pH value, short-time ultraviolet radiation, heat stress and the concentration of Tween-80 on the conidia germination of Isaria fumosorosea1 IF-1106. The results showed that the optimum temperature for conidial germination was 26 ℃, the range of suitable temperature was 23 ~32 ℃, and the conidia germinated at a high relative humidity, when RH lowers than 85%,the conidia were ungerminative. The conidia germination rate was high under the dark, and the conidia germination rate was low under the light. Under the condition of alternating light and dark, the rate has a trend of growing with the longer light time. And the optimal pH was 5~7, 0.05%~1% Tween-80 was used to prepare the suspension. With the treatment of heat stress and short-time ultraviolet radiation, the strong inhibition in conidia germination of Isaria fumosorosea1 was exhibited. So the best condition for conidia germination was 26 ℃,RH higher than 95%, pH6 and dark.
