益气化瘀解毒方治疗内毒素致ARDS大鼠的炎症作用研究

目的探讨益气化瘀解毒方（以下简称＂复方＂）对内毒素（lipopolysaccharide,LPS）诱发大鼠急性呼吸窘迫综合征（acute respiratory distress syndrome,ARDS）的保护作用及对炎症因子的影响。方法 50只雄性健康SD大鼠随机等分为5组,每组10只。5组大鼠按动物饲养标准进行适应性喂养1周。中药低、中、高剂量组,分别给予复方低、中、高剂量灌胃,每天1次,模型组及空白对照组每天予蒸馏水灌胃,每天灌胃量为1 m L/100 g,除空白对照组外其余4组7天后尾静脉注射LPS（2 mg/kg）造模,空白对照组及造模4组16小时麻醉下处死。取肺组织及腹主动脉血,进行大鼠肺大体病理、病理组织光镜观察,并检测血清中肿瘤坏死因子α（tumor necrosis factor alpha,TNF-α）、白细胞介素1β（interleukin 1beta,IL-1β）、IL-6及肺组织中p65蛋白含量。结果与空白对照组比较,模型组TNF-α升高（P〈0.05）。与模型组比较,低剂量组大鼠血清中TNF-α无明显降低（P〉0.05）,中药中、高剂量TNF-α水平显著降低（P〈0.05）,但高、中剂量组之间无明显差距（P〉0.05）。与空白对照组比较,模型组IL-1β升高（P〈0.05）;与模型组比较,低剂量组IL-1β降低（P〈0.05）,中与高剂量组IL-1β水平明显降低（P〈0.05）。与空白对照组比较,模型组IL-6升高（P〈0.05）;与模型组比较,低剂量组IL-6明显降低（P〈0.05）,中剂量组IL-6水平显著降低（P〈0.05）,高剂量组IL-6水平明显降低（P〈0.05）。与空白对照组比较,模型组p65升高（P〈0.05）;与模型组比较,低剂量组p65明显降低（P〈0.05）,中剂量组p65水平显著降低（P〈0.05）,中药高剂量组p65水平降低（P〈0.05）。结论益气化瘀解毒复方能够减轻内毒素致ARDS大鼠肺组织损伤,具有一定的预防保护作用,可能与降低大鼠血清中TNF-α、IL-1β、IL-6水平,抑制NF-κB通路激活,减轻炎症反应有关。在一定范围内,提?

Objective To study protective effect of Yiqi Huayu Jiedu recipe on acute respiratorydistress syndrome（ ARDS） rats induced by lipopolysaccharide and to observe the influence of inflammatory factor. Methods 50 healthy male SD rats were randomly divided into 5 groups（ n = 10）. All rats were fed for one week to adapt the environment. The rats were intragastric administration with correspondingdrug. The ARDS model was injected with LPS（ 2mg / kg） after 7 days. The rats were sacrificed after 16 hours of modeling. Lung tissue was collected to observed the pathology organization,and the TNF-α,IL-1β,IL-6,p65 protein in serum were detected. Results TNF- alpha： compared with the blank control group,serum TNF-α of rats in the model group was increased（ P〈0. 05）. Compared with the model group,TNF-α of low dose of Chinese medicine group was decreased non-significantly,TNF-α of medium and high dose of Chinese medicine group was decreased significantly（ P〈0. 01）,but there was no significant between two groups; Compared with the blank control group,IL-1,IL-6 and p65 protein of model group rats was increased（ P〈0. 05）. Compared with the model group,IL-1,IL-6 and p65 protein of Chinese medicine groups were decreased（ P〈0. 05）. Conclusion Yiqi Huayu Jiedu recipe can reduce the lung tissue injuries of ARDS rats induced by LPS,and can reduce the level of TNF-α,IL-1β,IL-6,can inhibit the NF-κB pathway activation and reduce inflammation reactions. In a certain range,improve the dosage of compound,and enhance the effect.
