采用反相乳液法制备了一种新型的自固化磷酸钙微球,其粒径范围为100~1000μm,中位径在388~605μm之间,内部有43.4%~66.7%的凝胶孔和微孔.通过正交试验获得了稳定制备微球的优化工艺参数:搅拌速度150r/min,明胶质量浓度8.0g/mL,油水相体积比10∶1,液固比2.0∶1.文中还研究了在此工艺条件下制备的自固化磷酸钙微球的流动性、力学性能、抗崩解性及细胞相容性.结果表明:所制备微球的流动性良好,自然堆积安息角为28.4°~31.6°;微球有良好的韧性,不易破碎,经过充分水化后,微球的抗压性能显著提高;微球在模拟生理条件下具有良好的抗崩解性和良好的细胞相容性.
In this work,a new type of self-setting calcium phosphate microspheres,the diameters and the medium diameter of which respectively ranged in 100~1 000 μm and 388~605 μm,was prepared via the reversed emulsion method,with a porosity of gel pores and micropores varying from 43.4% to 66.7%.The preparation parameters were optimized by orthogonal experiments,that is,an agitating speed of 150 r/min,a gelatin content of 8.0 g/mL,an oil-to-water ratio of 10∶ 1 and a liquid-to-powder ratio of 2.0 ∶ 1.Finally,the flowability,mechanical property,anti-disintegration and cytocompatibility of the microspheres prepared in the optimal conditions were investigated.The results indicate that the microspheres are of high flowability and toughness,with an angle of repose being up to 28.4°~31.6°,that the compressive performance of the microspheres significantly improves after a full hydration,and that the microspheres are endurable and biocompatible in simulated physiological conditions.
