目的:探讨树舌多糖GF辅助siRNA干扰沉默CDK2基因表达的作用。方法:本实验采用人肝癌细胞SMMC7721为研究对象,将中药树舌多糖GF2.5μg·mL-1与siRNA干扰序列191联合应用,采用Real-timePCR方法检测CDK2基因mRNA水平表达情况。结果:树舌多糖GF2.5μg·mL-1与191序列合用比单纯应用191序列抑制率提高3%。结论:树舌多糖GF2.5μg·mL-1与191序列合用比单纯应用191序列抑制率提高3%,表明树舌多糖GF对RNAi技术沉寂肝癌CDK2基因有一定的辅助作用。
Objective:To study the effect of[Polysaceharides component GF extracted by Ganoderma applanatum (Per. ex Gray) ] assisted siRNA interference silence CDK2 gene expression. Methods: In this experiment, human hepatocellular carcinoma cells line ( SMMC7721 ) used as the research object, the traditional Chinese medicine GAPSGF 2.5μgomL-1 and siRNA interference sequence 191 was applied and was detect the level of CDK2 gene expression of mRNA. with Real- time PCR method Results: Comparing with application of siRNA segment 191 alone,the inhibition ratio can? increase 3% by use GAPSGF and siRNA segment 191 combined ap- plication, Conclusions: Comparing with application of siRNA segment 191 alone, the inhibition ratio can increase 3 % by GAPSGF and siRNA segment 190 combined application,whick demonstrates that GAPSGF has certain assistant action for the RNA interfere expres- sion of CDK2.
