对松浦镜鲤口灌甲砜霉素,分别设对照组、50 mg/kg(1倍正常用药量,推荐剂量组)、150 mg/kg(3倍正常用药量,中等剂量组)及250 mg/kg(5倍正常用药量,毒性剂量组),连续口灌给药15 d,每天1次,于停药24 h后,自实验鱼的肝、脾、肾、肠和鳃采取新鲜组织,浸泡于波恩氏液中,待进行石蜡切片制作。结果显示,与对照组相比,松浦镜鲤肝、脾和肾在1倍给药剂量时,出现大量深染的炎性细胞,随着给药剂量增加,组织结构遭到破坏程度加重;3倍正常用药量时,肝、脾和肾出现细胞空泡化现象,肾小管管腔界限不清;在5倍正常用药量时,各组织空泡化现象加重,肝和脾出现细胞减少,细胞核溶解。对肠组织而言,1倍给药组与对照组无差异;3倍正常用药量时,肠绒膜出现孔隙;5倍正常用药量时,少量肠绒毛结构不完整,出现脱落现象。鳃组织与对照组相比,给药组出现鳃丝肿胀现象,随给药剂量加大,鳃丝肿胀加重。研究表明,在将甲砜霉素应用于松浦镜鲤的疾病防控时,应严格遵照药物的使用剂量进行给药,如果因病情原因或使用错误而造成使用剂量加大时,应对松浦镜鲤肝、肾等组织进行检查,来确定药物对鱼体的损伤程度。[中国渔业质量与标准,2014,4(3):54-59]
The study set the control group, 50 mg/kg (one time normal dose, the recommended dose group ), 150 mg/kg (three times normal dose, the medium dose group) and 250 mg/kg (five times normal dose, the toxic dose group) respectively. Thiamphenicol was administered 15 d consecutively on Songpu mirror carp( Cyprinus carpio L. ) through mouth one time per day. We collected fish tissues, including liver, spleen, kidney, intestine and gill severally 24 h after drug suspension, soaked them in Bonn's liquid for making the paraffin section. The results showed that compared with the control group, a large number of inflammatory cells occurred in liver, spleen and kidney of Songpu mirror carp in one time normal dose group. As the dose increased, the tissue structure was increasingly dam- aged. When it came to three times, liver, spleen and kidney cells demonstrated vacuolation, and renal tubular bound- ary was not clear. Liver, spleen and kidney ceils decreased and cell nucleus dissolved in five times dose group. Be-sides, cell vacuolation got worse. Intestinal tissue had no difference with control group in one time dose group. Holes occurred in intestinal chorion in three times dose group. In five times dose group, a few intestinal villus structures were incomplete and obscission appeared. As for the gill tissue, gill filaments appeared swollen in medication admin- istration groups compared with control group. And this phenomenon got aggravated as the dose increased. The study showed that when thiamphenicol is applied to disease control and prevention for Songpu mirror carp, drug should be strictly administered. If the dose is increased due to illness condition or operational error, the liver and kidney of Songpu mirror carp should be checked to determine its damage to the fish.
