目的:探讨藏雪莲水提取物能否减轻中波红斑效应紫外线(UⅤB)辐射后人角质形成细胞(HaCaT 细胞)的氧化损伤。方法体外培养 HaCaT 细胞,分为低剂量辐射组(30 mJ/ cm2,辐射1 h)、高剂量辐射组(60 mJ/ cm2,辐射1 h),低剂量辐射组进一步分为对照组、L - UⅤB组、L - UⅤB +低剂量臧雪莲组(臧雪莲1 g/ L)、L - UⅤB +高剂量臧雪莲组(臧雪莲5 g/ L),高剂量辐射组进一步分为对照组、H - UⅤB组、H - UⅤB +低剂量臧雪莲组(臧雪莲1 g/ L)、H - UⅤB +高剂量臧雪莲组(臧雪莲5 g/ L)。藏雪莲水提取物于 UⅤB 辐射前1 h 加入细胞培养基中,照射后18 h,收集细胞。通过台盼蓝染色观察细胞形态及死亡情况,MTS 比色法检测细胞增殖情况,选出氧化损伤较轻组行酶标法检测超氧化物歧化酶(SOD)活力、谷胱甘肽(GSH)含量、丙二醛(MDA)含量、过氧化氢酶(CAT)活力。结果 L - UⅤB组吸光度(A)值较对照组降低(P <0.05),L - UⅤB +低剂量臧雪莲组、L - UⅤB +高剂量臧雪莲组 A 值较L - UⅤB组升高(P <0.05);H - UⅤB组 A 值较对照组降低(P <0.05),H - UⅤB +低剂量臧雪莲组、H - UⅤB +高剂量臧雪莲组 A 值较H - UⅤB组升高(P <0.05)。L - UⅤB组 SOD 活力、GSH 含量、CAT 活力较对照组降低,MDA 含量较对照组升高(P <0.05);L - UⅤB +低剂量臧雪莲组 GSH 含量较L - UⅤB组降低、MDA 含量较L - UⅤB组升高(P <0.05);L - UⅤB +高剂量臧雪莲组 SOD 活力、GSH 含量、CAT 活力较L - UⅤB组和L - UⅤB +低剂量臧雪莲组升高,MDA 含量较L - UⅤB组和L - UⅤB +低剂量臧雪莲组降低(P <0.05)。结论低剂量藏雪莲水提取物对 UⅤB 辐射后 HaCaT 细胞无抗氧化损伤作用,高剂量藏雪莲水提取物对 UⅤB 辐射后 HaCaT 细胞有抗氧化损伤作用。
Objective To research whether Tibetan Saussurea water extracts could reduce the oxidative damage of UⅤB radiation on HaCaT cells. Methods HaCaT cells were cultured in vitro. The cultured cells were divided into low dose radiation group(30 mJ/ cm2 ,radiation 1 h),and high dose radiation group(60 mJ/ cm2 ,1 h). The low dose radiation group was further divided into control group,L - UⅤB group,L - UⅤB + low - dose TS group(TS 1 g/ L)and L - UⅤB + high - dose TS group(TS 5 g/ L). The high dose radiation group was further divided into control group,H - UⅤB group,H - UⅤB + low -dose TS group( TS 1 g/ L)and H - UⅤB + high - dose TS group( TS 5 g/ L). One hour before UⅤB radiation,Tibetan Saussurea water extracts were added into the culture medium,and 18 hours after radiation began,cells were collected. The morphology and death of cells were observed by trypan blue staining,and cell proliferation was detected by MTS. Selected the group with lighter oxidative damage to test SOD activity,GSH content,MDA content and CAT activity by enzyme linked immunosorbent assay. Results The A value in L - UⅤB group was significantly lower than that in control group,respectively(P〈 0. 05);the A value in L - UⅤB + low - dose TS group and L - UⅤB + high - dose TS group was significantly higher than that in L - UⅤB group,respectively(P 〈 0. 05). The A value in H - UⅤB group was significantly lower than that in control group, respectively(P 〈 0. 05);the A value in H - UⅤB + low - dose TS group and H - UⅤB + high - dose TS group was significantly higher than that in H - UⅤB group,respectively( P 〈 0. 05). The SOD activity,GSH content and CAT acivity in L - UⅤB group was significantly higher than that in control group;the MDA content in L - UⅤB group was significantly lower than that in control group,respectively(P 〈 0. 05). The GSH content in L - UⅤB + low - dose TS group was significantly lower than that in L - UⅤB group;the
