老鼠簕多糖的分离纯化及结构性质的研究

热水浸提法得到的老鼠簕粗多糖经过D301和三氯乙酸脱色脱蛋白后,利用DEAE-纤维素离子交换柱和Sepharose CL-6B凝胶柱层析纯化,得到两种多糖-老鼠簕中性多糖(NAP,neutral Acanthus ilicifolius L.polysaccharide)和老鼠簕酸性多糖B组分(AAPB,acid Acanthus ilicifolius L.polysaccharide B)。高效凝胶过滤色谱法(HPGFC)测得NAP、AAPB的分子质量分别为11775、23161。采用气相色谱、红外光谱和液相色谱仪等手段分析多糖的结构和组成成分。结果表明NAP主要由阿拉伯糖、葡萄糖和半乳糖组成,不含蛋白质。AAPB主要是由鼠李糖、阿拉伯糖和半乳糖组成。AAPB糖醛酸含量为51.23%。蛋白质含量为4.07%,是一种酸性果胶类杂多糖。

Crude polysaccharides extracted from Acanthus ilicifolius L. by hot water were decoloured and deproteinated by D301 and trichloroacetic acid. Two polysaccharide flactions named NAP(neutralAcanthus ilicifolius L. polysaccharide) and AAPB (acid Acanthus ilicifolius L. polysaccharide B)were isolated by anion-exchange chromatography DEAE-cellulose and size-exclusion chromatography of Sepharose CL-6B. The purified NAP and AAPB were proved to he homogeneous by HPGFC and their molecular weights (MW) were 11775 and 23161 respectively. IR,CJC and LC were used to explore structural characters of the polysaccharides. Analysis of neutral monosaccharide composition indicated that NAP contained high proportions of arabinose, galactose and glucose, and AAPB was mainly composed of rhamnose, arahinose and galactose. The content of uronic acid in AAPB was 51.23% and that of protein in AAPB was 4.07%, which showed that AAPB is a pectin-type heteropolysaccharide.
