从江苏省泰州市姜堰地区某鹅场采集小鹅瘟疑似病例,采用鹅胚接种、血凝、琼脂扩散、PCR检测、动物感染、冰冻切片间接免疫荧光等试验对病原进行鉴定。结果显示,所分离的毒株为鹅细小病毒,动物回归试验中雏鹅感染病毒后出现与自然病例相似的症状,且该病毒具有很强的致病性,ELD50为10-5.9/0.2mL,LD50为10-4.1/0.3mL,扩增的VP3基因测序结果与其他鹅细小病毒毒株比较具有很高的同源性,且与鹅细小病毒标准B毒株的VP3基因序列同源性为96.5%。分离的病毒经鉴定为鹅细小病毒强毒株。
A suspected case of the disease GPV was collected from a goose farm in Taizhou Jiangyan city of Jiangsu province.The inoculation of goose embryos,hemagglutination(HA),agar diffusion(AGP),PCR testing,animal infection and indirect immunoinfluscent assay(IFA)tests were used to identify the isolated virus.The tests showed that the isolated virus was GPV.And the animal regression test also verified that geese had the similar symptoms to those found in the field,and it had strong pathogenicity.ELD50 was 10-5.9/0.2 mL,and LD50 was 10-4.1/0.3 mL.VP3 gene amplification and sequencing results showed that it had high homology with GPV,and the VP3 gene homogeneity with B standard strain was 96.5%.The isolated virus was identified as GPV with strong pathogenicity.
