选用3株新城疫病毒(NDV)分离株(鸡源JSC0804、鸭源JSD0812、鹅源JSG0210)及标准毒株F48E8感染鸡胚成纤维细胞,经3次蚀斑纯化后,获得4株NDV的纯化株。对纯化毒株的MDT、ICPI、TCID50、pfu等毒力指标进行测定,并将4个毒株以相同的病毒细胞感染复数(MOI)感染鸡、鸭和鹅胚成纤维细胞。结果表明,纯化后毒株较原毒株毒力有所增强,4个毒株都能在3种成纤维细胞上稳定产生细胞病变,但鸭源NDV分离株JsD0812对鸭胚成纤维细胞感染性及鹅源NDV分离株JSG0210对鹅胚成纤维细胞感染性均分别较其他3个毒株强。
Three virulent Newcastle disease virus (NDV) field strains, chicken origin strain JSC0804, duck origin strain JSD0812 and goose origin strain JSG0210, and standard virulent strain F48E8 were plaque-pu- rified three times on chicken embryo fibroblasts. The virus titers and virulence indexes such as median tis- sue culture infective dose (TCIDs0), plaque forming unit (pfu), mean death time (MDT), and intracere- bral pathogenicity index (ICPI) of purified strains were monitored, and chicken embryo fibroblasts (CEF), duck embryo fibroblasts (DEF) and goose embryo fibroblasts (GEF) were infected with the four strains at the same multiplicity of infection (MOI), respectively. The results showed that virulences of pu- rified strains were increased compared to those of the original ones. All four purified strains can produce stable cytopathic effect (CPE) in three kinds of avian fibroblasts, but there was a little difference in CPE among strains. The infectivity of duck origin strain JSD0812 to DEF, and goose origin strain JSG0210 to GEF was higher than that of other three strains, respectively.
