目的了解血小板特异性抗原(HPA)-1—6、15基因多态性在拉萨藏族献血人群中的分布特征。方法.采用聚合酶链式反应.序列特异性引物(PCR—SSP)技术对405名藏族无关献血者作HPA-1—6、15基因分型。络果405名无关献血者HPA各等位基因的频率分别为:HPA—1a:0.9383,HPA-16:0.0617;HPA-2a:0.9716,HPA-2b:0.0284;HPA.3a:0.6173,HPA-3b:0.3827;HPA-4a:0.9926,HPA-4b:0.0074;HPA-5a:0.9494,HPA-5b:0.0506;HPA.6a:0.9914,HPA-6b:0.0086;HPA-15a:0.6037,HPA-15b:0.3963。人群资料比对结果显示,与南方汉族人群(n=1554)相比,拉萨藏族献血人群HPA-1、HPA-2、HPA-3、HPA.-4、HPA-5、HPA-15系统基因频率具有明显差异;与新疆柯尔克孜族人群(n=105)相比,仅HPA-2系统基因频率具有明显差异。拉萨藏族献血人群HPA-1b等位基因频率明显低于高加索人群(n=119),而与新疆柯尔克孜族人群(n=105)相近,但明显高于亚洲其他黄种人群。结论拉萨藏族献血人群的HPA各位点基因频率显示出明显的民族和地域性特征。
Objective To investigate the allele distribution of HPA-1 - 6, 15 in Titetan blood donors from Lha- sa. Methods The genotyping of HPA-1 - 6,15 were performed by the PCR with sequence-specific primers (PCR-SSP). A total of 405 healthy volunteer unrelated Tibetan blood donors were included. Results The allele frequencies of HPAs for 405 Tibetan individuals were HPA-la: 0. 9383,HPA-lb: 0.061 7 ;HPA-2a: 0. 97i 6,HP.A-2b: 0. 028 4; HPA-3a: 0. 617 3, HPA-3b : 0. 382 7 ;HPA-4a: 0. 992 6 ,HPA-4.b : 0. 007 4.; HPA-Sa: 0. 949 4 ,HPA-5 b : 0. 050 6 ; HPA-6a : 0. 991 4, HPA-6b : 0. 008 6 ;HPA-15a: 0. 603.7 ,HPA-15b : 0. 396 3 ,respectively. The comparison with data of other ethnic groups showed that the allele frequencies of HPA-1 ,HPA-2,HPA-3 ,HPA.4,HPA-5 and HPA-15 were all significantly different between Tibetan and South Chinese Han( n = 1 554). While compared with Xinjiang Khalkhas ( n = 105), such a significant difference was only found in HPA-2 system. The HPA-lb allele frequency in the Tibetan was significantly lower than that found in Caucasian from Germany ( n = 119 ), but signiiicantly higher than the other compared Mongolian race; except for Xinjiang Khal- khas. Conclusion Our study demonstrates that the distribution of HPA-1 - 6,15 in Tibetan exhibit unique characteristics.
