目的探讨着色性干皮病基因D(XPD)的单核苷酸多态性(SNPs)与铂类药物化疗敏感性的关系。方法采用基因测序法对50例经病理确诊且CT扫描证实有可测量病灶的恶性肿瘤患者的外周血进行XPD基因SNPs检测,分析各基因型与化疗敏感性的关系。结果 XPD基因型的分布与病种无关(P〉0.05)。XPD基因第312位密码子的纯合突变型Asn/Asn基因分型对铂类化疗的敏感性高于纯合野生型Asp/Asp及杂合突变型Asp/Asn基因分型(P〈0.05)。结论 XPD基因SNPs可能与铂类药物化疗敏感性有关。
Objective To investigate the relationship between single nucleotide polymorphisms(SNPs) of xeroderma pigmentosum complementation group D(XPD) and sensitivity of platinum based chemotherapy.Methods Direct sequencing was performed to detect XPD genotypes in peripheral blood from 50 patients with cancer receiving platinum based chemotherapy.Results Homozygous mutant Asn/Asn genotype had higher response rate than wild type Asp/Asp and heterozygous mutant Asp/Asn genotype in XPD 312 codon(P0.05).Conclusion SNPs of XPD gene may be associated with the sensitivity of platinum based chemotherapy.
