目的联合多家采供血机构开展全血制备浓缩血小板的汇集及滤除白细胞的研究,为制定全血制备浓缩血小板的汇集及滤除白细胞的操作规程和质量标准提供依据。方法采用PRP法或BC法由400ml新鲜全血制备浓缩血小板,将10—16UABO同型的浓缩血小板汇集,随机用A、B两种国产血小板型去白细胞滤器进行过滤,评价过滤前后血小板的质量和功能,共完成202例研究。结果汇集血小板数≥2.4×10^11个的共147例,其中A组77例,B组70例。A组过滤前后的血小板数、白细胞数分别为(2.90±0.45)×10^6vs(2.60±.43)×10^11,(176.45±135.67)×10^6vs(1.00±2.29)×10^6;B组过滤前后的血小板数、白细胞数分别为(2.80±0.36)×10“^6vs(2.40±0.37)×10^11,(175.76±147.84)×10^6vs(0.30±0.72)×10^6汇集血小板数〈2.4×10^11个的共55例,其中A组29例,B组26例。A组过滤前后的血小板数、白细胞数分别为(1.71±0.39)×10vs(1.43±0.42)×10^11,(65.85±110.97)×10^6vs(3.7±3.98)×10^6;B组过滤前后的血小板数、白细胞数分别为(1.79±0.48)×10^6vs(1.42±0.46)×10^11,(70.63±145.55)×10^6vs(1.45±2.66)×10^6。A、B两组过滤前后pH值、血小板低渗休克、CD62p阳性表达率和聚集率无显著差异。结论对PRP法或BC法制备的浓缩血小板进行汇集、过滤,可有效去除白细胞,且过滤前后血小板功能无显著变化,符合相关标准的要求。本研究为制定全血制备浓缩血小板的汇集及滤除白细胞的操作规程和质量标准提供了依据。
Objective To cooperate with a number of blood centers and evaluate the quality of pooled platelet and leu-kocyte depletion, which aims to supply basis for establishing operating procedures and quality standard of pooled platelet and leukocyte depletion. Methods PCs was prepared from 400 ml fresh whole bood by platelet-rich plasma (PRP) or buffy- coat (BC) method. 10 to 16 units of ABO-matched PCs were pooled,and the. filtered with two types of domestic filters (namely group A and B) randomly. The conventional and functional indicators of platelet were evaluated before and after filtering, and the total samples were 202. Results The total of cases that number of platelet was more than 2. 4 ×10^11 was 147 ,including 77 cases in group A and 70 cases in group B. In group A the number of platelet and leukocyte before and af- ter filtering were(2.90 ±0. 45) ×10^11 VS(2.60 ±0. 43) ×10^11 , ( 176.45 ± 135. 67) ×10^6 vs( 1. 00 ±2.29) ×10^6 respec- tively,and in group B the number of platelet and leukocyte before and after filtering were(2. 80 ±0. 36) ×10^11vsS(2. 40± 0. 37 ) ×10^11 , ( 175.76 ± 147. 84) ×10^6 VS(0. 30 ± 0. 72) ×10^6 respectively. While the total of cases that number of plate- let was less than 2.4 ×10^11 was 55 ,including 29 cases in group A and 26 cases in group B. In group A the number of plate- let and leukocyte before and after filtering were( 1.71±0. 39) ×10^11 VS( 1. 43 ±0. 42) ×10^11 , (65.85 ± 110. 97) ×10^6 VS (3.7 ±3.98) ×10^6 respectively,and in group B the number of platelet and leukocyte before and after filtering were( 1.79 ± 0. 48) ×10^11 VS( 1.42 ±0. 46) ×10^11, (70. 63 ±145.55) ×10^6 VS( 1.45 ± 2. 66) ×10^6 respectively.
