目的观察艾灸对大鼠腹膜透析结构与功能的影响,探讨艾灸疗法辅助腹膜透析临床应用的实验基础。方法采用腹腔注射腹透液制备大鼠腹透模型,随机分为正常组、模型组、假透组、艾灸组,其中艾灸组以腹腔注射前30 min艾灸大鼠腹部神阙穴5 min为治疗方案。实验完毕,大鼠行腹膜平衡试验,记录腹膜超滤量,采用化学比色法测定大鼠腹膜物质清除率、透析液溶质浓度与血液溶质浓度(D/P)比值,观察艾灸对腹膜功能的影响。结果与正常组相比,模型组大鼠腹膜超滤量减少(P〈0.05),葡萄糖D/P比值降低(P〈0.05),肌酐、尿素氮清除率,K+、Na+清除率及D/P比值(肌酐、尿素氮)有降低趋势,统计无显著性差异;与模型组相比,艾灸组大鼠腹膜超滤量增加(P〈0.05),尿素氮清除率及D/P比值(肌酐)提高(P〈0.05),葡萄糖D/P比值增加(P〈0.05),肌酐清除率,K+、Na+清除率及D/P(肌酐、尿素氮)也有增加趋势,统计无显著性差异。结论艾灸腹部神阙穴能一定程度上改善大鼠腹膜功能。
Objective To investigate the effect of moxibustion on rat peritoneal membrane structure and osmosis and explore the experimental basis for the clinical application of moxibustion-assisted peritoneal dialysis.Methods A rat model of peritoneal dialysis was made by intraperitoneal injection of dialysate.The rats were randomly allocated to normal,model,sham dialysis and moxibustion groups.In the moxibustion group,5 min moxibustion on rat abdominal point Shenque was performed at 30 min before intraperitoneal injection.At the end of experiment,a peritoneal equilibration test was conducted,the dialysate was drained from the abdominal cavity and abdominal aortic blood was taken in the rats.Rat peritoneal substance clearance rate and the ratio between dialysate and blood solute concentrations(D/B) were determined by chemical colorimetry and double antibody-sandwich enzyme linked immunosorbent assay,respectively,to investigate the effect of moxibustion on the peritoneal function.Results As compared with the normal group,peritoneal ultrafiltration volume and glucose D/B ratio decreased(both P〈0.05),and creatinine and urea nitrogen clearance rates,K+ and Na+ clearance rates and D/B ratios(creatinine and urea nitrogen) tended to decrease,which had no statistically significant differences,in the model group of rats.As compared with the model group,peritoneal ultrafiltration volume,urea nitrogen clearance rate,D/B ratio(creatinine) and glucose D/B ratio increased(all P〈0.05),and creatinine clearance rate,K+ and Na+ clearance rates and D/B ratio(urea nitrogen) tended to increase,which had no statistically significant differences,in the moxibustion group of rats.Conclusion Moxibustion on abdominal point Shenque can improve rat peritoneal membrane structure and osmosis to a certain extent.
