目的研究隔药灸与电针大肠下合穴上巨虚对溃疡性结肠炎(UC)大鼠结肠黏膜肠三叶因子(ITF)、白介素8(IL-8)表达的调节作用。方法将大鼠随机分为正常组、模型组、隔药灸组、电针组。采用免疫法加局部刺激制备UC大鼠模型,隔药灸组、电针组选取上巨虚穴,分别进行隔药灸、电针治疗,连续治疗14次。治疗后记录大鼠症状体征变化;光镜下观察结肠形态学改变;采用免疫组化观察ITF、IL-8的表达变化。结果溃疡性结肠炎大鼠结肠黏膜ITF表达显著低于正常组(P〈0.01),IL-8表达明显高于正常组(P〈0.05);隔药灸组、电针组治疗后,大鼠体重增加,腹泻、脓血便等症状体征和结肠形态学损伤均明显改善;结肠黏膜ITF表达均较模型组显著增强(P〈0.01),IL-8表达均较模型组明显降低(P〈0.05)。结论UC大鼠结肠黏膜ITF表达明显降低,IL-8表达显著增高,隔药灸、电针上巨虚穴均能调节ITF和IL-8的异常表达。
Objective To investigate the regulating effects of medicinal paste moxibustion and electroacupuncture on intestinal trefoil factor (ITF) and interleukin-8 (IL-8) expressions in the colonic mucosa of ulcerative colitis (UC) rats. Methods Rats were randomly allocated to normal, model, medicinal paste moxibustion and electroacupuncture groups. A rat model of UC was made by an immunological method plus local stimulation. Point Shangjuxu was selected for medicinal paste moxibustion and electroacupuncture in the medicinal paste moxibustion and electroacupuncture groups. The treatments continued for 14 times. After treatment, changes in the rat’s symptoms and signs were recorded, changes in colonic morphology were observed under an optical microscope, and ITF and IL-8 expressions were examined by an immunohistochemical method. Results ITF expression decreased obviously (P〈0.01) and IL-8 expression increased obviously (P〈0.05) in the colonic mucosa of ulcerative colitis rats compared with the normal group. After treatment, body weight increased, the symptoms and signs such as diarrhea and bloody purulent stool improved, and morphological injuries to the colon were reduced in the medicinal paste moxibustion and electroacupuncture groups. ITF expression increased obviously (P〈0.01) and IL-8 expression decreased obviously (P〈0.05) in the colonic mucosa of both the medicinal paste moxibustion and electroacupuncture groups compared with the model group. Conclusion ITF expression decreased obviously and IL-8 expression increased obviously in the colonic mucosa of UC rats. Both medicinal paste moxibustion and electroacupuncture at point Shangjuxu can regulate abnormal ITF and IL-8 expressions.
