目的研究温和灸对自然衰老大鼠肝组织Rb、P53、Bcl-2、PKC蛋白及其mRNA表达的影响。方法 24只SD大鼠随机分为正常组、自然衰老组、艾灸组。正常饲养20个月,待其自然进入衰老期以制备自然衰老大鼠模型,温和灸自然衰老大鼠双侧肾俞穴对其进行干预。应用原位杂交、免疫组化方法检测各组大鼠肝组织Rb、P53、Bcl-2、PKCmRNA及其蛋白表达。结果与正常组比较,自然衰老组大鼠肝组织Rb、P53mRNA及蛋白表达均显著增强(P〈0.01,P〈0.05),Bcl-2、PKCmRNA及蛋白表达均显著降低(P〈0.01,P〈0.05);与自然衰老组大鼠比较,艾灸组大鼠肝组织Rb、P53mRNA及蛋白表达均显著降低(P〈0.01,P〈0.05),Bcl-2、PKCmRNA及蛋白表达均显著增强(P〈0.01,P〈0.05)。结论温和灸肾俞穴能降低自然衰老大鼠肝组织Rb、P53mR-NA及蛋白表达,增强Bcl-2、PKCmRNA及蛋白表达。
Objective To explore the effect of warm moxibustion on the expression of Rb,P53,Bcl-2 and PKC in liver tissue of natural aging rats.Methods 24 SD rats were randomly assigned into normal group,natural aging group and warm moxibustion group.The model of natural aging rats was established by being normally bred for twenty months.The rats in warm moxibustion were treated by warm moxibustion at Shenshu.The expression of Rb,P53,Bcl-2 and PKC was measured by in situ hybridization and immunohistochemistry.Results Compared with normal group,the mRNA and protein expression of Rb and P53 were significantly increased(P〈0.01,P〈0.05) and those of Bcl-2 and PKC were significantly decreased(P〈0.01,P〈0.05) in natural aging rats.Compared with natural aging group,the mRNA and protein expression of Rb and P53 were significantly decreased(P〈0.01,P〈0.05) and those of Bcl-2 and PKC were significantly increased(P〈0.01,P〈0.05) in warm moxibustion group.Conclusions Moxibustion at Shenshu can decrease the mRNA and protein expression of Rb and P53,and increase the mRNA and protein expression of Bcl-2 and PKC in liver tissue of natural aging rats.
