目的:研究五加生化缓释片对功能性子宫出血的作用及机制。方法选取40只雌性KM小鼠,随机分成空白组、产复康颗粒组以及五加生化缓释片低、中、高剂量组,每组8只,采用断尾法和毛细管凝血法测定小鼠的出血及凝血时间。另取早孕SD大鼠30只,随机分成模型组(造模方法:灌服米非司酮和米索前列醇造成不完全流产模型)、产复康颗粒组以及五加生化缓释片低、中、高剂量组,每组6只,采用毛细玻璃管法测定出血时间,采用剪尾法测定凝血时间,采用酶标仪对置入阴道棉球的血浸渍液进行子宫出血量测定,观察五加生化缓释片对该模型大鼠出、凝血时间及子宫出血量的影响。结果与空白组比较,五加生化缓释片高、中、低剂量组能明显缩短小鼠的出血及凝血时间(P〈0.01、P〈0.05);而与产复康颗粒组比较,五加生化缓释片高、中剂量组能明显缩短小鼠的出血及凝血时间(P〈0.01);与空白组比较,五加生化缓释片高、中、低剂量组能明显减少大鼠的子宫出血量(P〈0.01、P〈0.05);与产妇康颗粒组比较,五加生化缓释片高剂量组能明显减少大鼠的子宫出血量(P〈0.01)。结论五加生化缓释片能显著减少大鼠的子宫出血量。
Objective To study the effect and mechanism of action of Wujia Shenghua sustained-release tablets in dysfunctional uterine bleeding. Methods 40 KM female mice were selected and divided into the blank group,the Chanfukang pellet group and Wujia Shenghua sustained-release tablets low,medium and high dose group,each group of eight,the bleeding and clotting time of mice were evaluated by tail and capillary blood coagulation method.Another 30 only pregnant SD rats were selected and divided into model group (building methods:to take mifepristone and misoprostol in incomplete abortion model),Chanfukang pellet group and Wujia Shenghua sustained-release tablets low,medium and high dose group,each group of six,the bleeding time was evaluated by capillary glass tube method,the clotting time was evaluated by cut tail method,the uterine blood loss used enzyme standard meter for determination of blood soaking liquid into the vagina tampon,the impact of clotting time and uterine blood loss of Wujia Shenghua sustained-release tablets in model rats were observed. Results Compared with the blank group,the Wujia Shenghua sustained-release tablets low,medium and high dose group could significantly shorten the bleeding and coagulation time in mice (P〈0.01, P〈0.05).But compared with Chanfukang pellet group,the Wujia Shenghua sustained-release tablets medium and high dose group could significantly shorten the bleeding and and coagulation time in mice (P〈0.01,P〈0.05).Compared with blank group,the Wujia Shenghua sustained-release tablets medium and high dose group could significantly shorten the rats uterus bleeding (P〈0.01,P〈0.05).Compared with Chanfukang pellet group,the Wujia Shenghua sustained-release tablets high dose group could significantly shorten the rats uterus bleeding (P〈0.01). Conclusion Wujia Shenghua sustained-release tablets can significantly re-duce the uterine blood loss in rat.
