目的观察α7烟碱样乙酰胆碱受体(α7 nicotinic acetylcholine receptor,α7n AChRs)对原代海马神经元存活率及突触可塑性的调节作用。方法将原代培养的海马神经元随机分为对照组(正常培养无任何干预);α7胆碱能受体拮抗剂作用1 h、3 h、6 h、12 h、24 h、48 h组。培养24 h的神经元换液后给予药物作用。作用终止后进行MTT代谢率和LDH漏出率检测。然后观察细胞模型突触的形态;Western blot检测神经颗粒素(Neurogranin,Ng)和神经调节素(Neuromodulin,Nm)的表达。结果 (1)MTT代谢率和LDH漏出率实验结果显示,相对于对照组,随着α7胆碱能受体拮抗剂作用时间延长其对海马神经元增殖的抑制作用更加明显;(2)在共聚焦显微镜下观察到,相对于对照组,α7胆碱能受体拮抗剂作用6 h后多突起神经元占总神经元的比例以及海马原代神经元突起长度明显低于对照组;(3)qRT-PCR和Western blot实验发现,相对于对照组,α7胆碱能受体拮抗剂作用6 h、12 h和24 h后Ng和Nm的表达均显著减少。结论 (1)α7胆碱能受体的拮抗剂明显抑制原代海马神经元的增殖及突触可塑性;(2)α7胆碱能受体的拮抗剂明显抑制原代海马神经元Ng和Nm的表达。
Objective To observe the effects of α7n AChRs on hippocampal neuron synaptic morphology and survival in vitro. Methods The primary cultured hippocampal neurons were divided into six groups,control group( remained in the incubator),methyllycaconitine( MLA) 1 h group、MLA 3 h group、MLA 6 h group、MLA 12 h group、MLA 24 h group、MLA48 h group. The neurons were stimulated after 24 h incubationor. The effects of MLA on neurons proliferation were observed by MTT and LDH assay. Then selected the appropriate time to AD cell model,synaptic morphology was observed by confocal microscopy,and the expressing of Ng and Nm were observed by Western blot. Results( 1) MLA inhibited of proliferation of hippocampal neurons.( 2) Synaptic morphology was inhibited by MLA.( 3) The expressingion of Ng and Nm were inhibited by MLA. Conclusion MLA significantly inhibited proliferation and synaptic plasticity of hippocampal neurons.
