[目的]研究DNA甲基转移酶1(DNMT1)对类风湿性关节炎(RA)模型大鼠关节滑膜成纤维样滑膜细胞(FLS)凋亡的影响。[方法]采用Real-timeqPCR检测对照大鼠和RA模型大鼠FLS中DNMT1表达,检测DNMT1siRNA转染对抗凋亡基因Bcl-2表达的影响;采用MTT检测DNMT1siRNA转染对FLS增殖的影响。[结果]与对照组相比,RA模型大鼠FLS中DNMT1表达显著升高,DNMT1表达抑制后,Bcl-2表达显著降低:M1Tr检测发现。DNMTI表达抑制后,FLS增殖活力显著减弱。[结论]DNMT1表达抑制可能引发RA模型大鼠FLS凋亡。
[ Objective ] The effects of DNA methyltransferase 1 (DNMT1) on the apoptosis of fibroblast-like synovial cells (FLS) in rheumatoid arthritis(RA)model rats were studied. [Method] The expression of DNMT1 in RA model rats and control rats was detected by using Real-time qPCR. And the effects of siRNA transfection of DNMT1 on the expression of anti-apoptosis gene Bcl-2 were studied. The effects of siRNA transfeetion of DNMT1 on the proliferation of FLS were detected by MTY. [Result] Compared with control group, the expression of DNMT1 in FLS of RA model rats was significantly up-regulated. After the expression of DNMT1 was inhibited,the expression of Bcl-2 was significantly down-regulated. The results of MTF detection showed that the proliferation activity of FLS was significantly reduced after the inhibition of DNMT1 expression. [ Conclusion ] The inhibition of DNMT1 expression might lead to the apoptosis of FLS in RA model rats.
